Although leading 4-1 in the first set and being an unseeded player in the US Open, Martincová was defeated 4-6, 0-6, by Victoria Azarenka, the 18th seed. In Ostrava, Martincová defeated Anastasia Pavlyuchenkova and Katerina Siniakova on the way to her maiden WTA 500 level quarterfinal before falling to eventual champion Maria Sakkari. She lost a heartbreaking match at Kremlin Cup to eventual 2021 WTA Finals champion Garbie Muguruza in the second round, falling 4-6, 6-4, 3-6. Muguruza is ranked No. 6 in the world. She debuted in the top 50 in November and finished the year ranked 48 in the world. Prior to the Australian Open, she only won one singles match, but she did make it to her first WTA doubles final at the Melbourne Summer Set 2 with Mayar Sherif and other doubles final the following week at the Adelaide International 2, where she lost both matches in three sets. After helping Vondrouová advance to the second round of the Australian Open, Martincová entered the top 100 for the first time in doubles.
